Principle of the Assay |
LTB4 ELISA kit applies the competitive enzyme immunoassay technique utilizing an anti-LTB4 antibody and an LTB4-HRP conjugate. The assay sample and buffer are incubated together with LTB4-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The result of the enzyme-substrate reaction is a complex that is blue in hue. The process is finally stopped by adding a stop solution, causing the solution to turn yellow. In a microplate reader, the color intensity is measured spectrophotometrically at 450 nm. Due to competition for the anti-LTB4 antibody binding site between LTB4 from samples and LTB4-HRP conjugate, the intensity of the color is inversely proportional to the concentration of LTB4. As more sites are taken up by LTB4 from the sample, fewer sites are left that can bind LTB4-HRP conjugate since the number of sites is restricted. A standard curve is drawn connecting the color's intensity (O.D.) and standard concentration. This standard curve is interpolated to determine the LTB4 concentration in each sample. |
