Principle of the Assay |
TESTO ELISA kit applies the competitive enzyme immunoassay technique utilizing an anti-TESTO antibody and an TESTO-HRP conjugate. The assay sample and buffer are incubated together with TESTO-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The result of the enzyme-substrate reaction is a complex that is blue in hue. The process is finally stopped by adding a stop solution, causing the solution to turn yellow. In a microplate reader, the color intensity is measured spectrophotometrically at 450 nm. Due to competition for the anti-TESTO antibody binding site between TESTO from samples and TESTO-HRP conjugate, the intensity of the color is inversely proportional to the concentration of TESTO. As more sites are taken up by TESTO from the sample, fewer sites are left that can bind TESTO-HRP conjugate since the number of sites is restricted. A standard curve is drawn connecting the color's intensity (O.D.) and standard concentration. This standard curve is interpolated to determine the TESTO concentration in each sample. |
